Several different sorts of clinical procedures have been developed to cancer
therapy during the last century. The classical cancer treatment includes the total excision of
tumor and adjacent tissues in combination with chemotherapy, immunotherapy or radiation
treatment. The development of hyperthermia has brought an additional driving force to
cancer therapy. In the hyperthermia treatment magnetic particles introduced in the tumors
are inductively heated by a magnetic field at moderate temperatures (41- 43 °C). The
temperatures that exceed 41 °C inactivate the cancer cells preserving the normal cells of the
surround tissues. Due to their biocompatible properties, poly (2-hydroxy
methylmethacrylate) (PHEMA) microspheres and nanospheres are among the most
promising carriers for the magnetic particles. An important part of our laboratory’s research
is focused on the encapsulation of the Y3Fe5-xA1xO12 (YFeAl) magnetic particles by using
biocompatible polymers for hyperthermia treatment. When the microencapsulated magnetic
materials are protected from extracellular enzymatic degradation and the citotoxicity of the
YFeAl due to metal presence may be avoided. Poly (2-hydroxy methylmethacrylate)
microspheres containing the polycrystalline particles of Y3Fe5-xAlxO12 (0≤x≤2) were
prepared by suspension polymerization in a batch reactor using ammonium persulfate and
as initiator and the cross-linker ethylene glycydil dimethacrylate EGDMA. The synthetized
microspheres were characterized by X-ray diffraction (XRD) and scanning electron
microscopy (SEM). The Curie temperature (Tc) was determined from magnetic
susceptibility measurements in the temperature rang of 223-573 K. The derivative curve for
the size of distribution determined by differential granulometric analysis and the SEM
micrograph for PHEMA/YFeAl microspheres showed good monodispersities (> 80%). The
XRD analysis reveals that yttrium aluminum iron garnet samples appear as a single phase.
SEM micrograph of YIG reveals the presence of aggregates of irregular fine particles. The
Curie temperatures (Tc) of YIG estimated from the magnetization curves decreased with
the aluminum content probably due to the reduction of the number of the main magnetic
interaction. The Tc values for the PHEMA microspheres containing YFeAl in the
composition range of 1.5≤x≤1.8 were near to room temperature, indicating that the
synthetized microspheres are promissory materials for the hyperthermia treatment. In order

to evaluate the possible influence of the PHEMA/YFeAl microspheres on cells, cytotoxicity
assay was carried out. It was observed that the presence of the microspheres did not affect
the cell viability or the culture growth rate. According to the results obtained in this work,
the synthetized PHEMA/YFeAl microspheres seems to be promising material for the
hyperthermia treatment. The PHEMA/YFeAl microspheres modulates the release of the 5-
Fluorouracil (5-FU) from magnetic microspheres, avoiding the high concentrations of the
drug that may cause severe systemic toxicity. Loaded PHEMA/YFeAl microspheres showed
a non-significant hyperemia, displaying low inflammatory reaction after implantation. The
results obtained in this work are promising according to the design of the drug delivery
systems in which is necessary to control both, the total amount of the drug and the kinetic
profile. |

Uma variedade de procedimentos clínicos tem sido desenvolvido para a terapia do
câncer durante o último século. O tratamento clássico do câncer inclui a retirada total do
tumor e tecidos adjacentes em combinação com a quimioterapia, imunoterapia ou
tratamento por radiação. O desenvolvimento da hipertermia trouxe um impulso adicional a
terapia do câncer. No tratamento através da hipertermia as partículas magnéticas
introduzidas nos tumores são aquecidas por indução através de um campo magnético em
temperatura moderada (41-43 °C). As temperaturas que excedem 41 °C inativam as células
cancerígenas preservando as células normais dos tecidos adjacentes. Devido as suas
propriedades biocompatíveis, as microesferas poli (2-hidroxi metilmetacrilato) (PHEMA) e
nanoesferas estão entre as mais promissoras transportadoras de partículas magnéticas. Uma
importante área de nossa pesquisa de laboratório está focalizada na encapsulação de
partículas magnéticas de Y3 Fe5-xAlxO12 (YFeAl) usando polímeros biocompatíveis para á
produção da hipertermia. Quando microencapsulados os materiais magnéticos são
protegidos de uma degradação enzimática extracelular e da citotoxicidade do YFeAl, devido
a presença do metal. As microesferas poli (2-hidroxi metilmetacrilato) contendo as
partículas policristalinas de Y3Fe5-xAlxO12 (0≤x≤2) foram preparadas pela polimerização da
suspensão num grupo de reatores usando persulfato de amônio e etileno glicidil
dimetacrilato EGDMA. As microesferas sintetizadas foram caracterizadas por difração de
raios-X (XRD) e microscopia eletrônica de varredura (SEM). A temperatura Curie (Tc) foi
determinada das medições de sensibilidade magnética nos limites de temperaturas de 223-
573 K. A curva derivada para o tamanho da distribuição determinada pela análise de
diferencial granulométrico e a micrografia SEM por microesferas PHEMA/YFeAl
mostraram monodispersibilidades (> 80%). A análise XRD revelou que as amostras de ítrio
alumínio ferro garnet apareceram como uma só fase. A micrografia SEM de YFeAl revelou
a presença de agregados de partículas finas irregulares. A temperatura Curie (Tc) de YFeAl
estimada das curvas de magnetização decresceu com o conteúdo de alumínio
provavelmente devido à redução do número de interação magnética principal. Os valores da
Tc para as microesferas PHEMA contendo YFeAl nos limites de composição de 1.5≤x≤1.8
estavam próximos a temperatura ambiente, indicando que as microesferas sintetizadas são

materiais promissores para o tratamento por hipertermia. A fim de avaliar a possível
influência das microesferas PHEMA/YFeAl nas células, testes de citotoxicidade foram
realizados. Foi observado que as microesferas não afetaram a viabilidade das células ou a
taxa de crescimento da cultura celular. Conforme os resultados obtidos neste trabalho, as
microesferas PHEMA/YFeAl sintetizadas parecem ser um material promissor para o
tratamento por hipertermia. As microesferas PHEMA/YFeAl modularam a liberação de 5-
Fluoruracil (5-FU) das microesferas magnetizadas, evitando as altas concentrações da
droga que pode causar severa toxicidade sistêmica. As microesferas carregadas
PHEMA/YFeAl mostraram uma hiperemia não significante, apresentando uma baixa reação
inflamatória após a implantação. Os resultados obtidos neste trabalho são promissores para
o projeto do sistema de fornecimento da droga no qual é necessário controlar ambos, a
quantidade total da droga liberada e o perfil cinético. |
